So, just to make sure, if you attack a fire-weak monster with a fire attack, it'll do 150% damage of what it does to regular monsters right? And if they are fire-strong, then it'll do 50%?
Does element weakness affect damage over time too?

You are correct when it comes to mages. It does affect DoT too.
I remember it used to work differently for archers.
I'm sure it works differently for paladin charges.

Paladin charges work the same way - 150% damage on elemental weak mobs, 50% on elemental resistant mobs. But the dual charging adds up the percents of the two skills, not multiplies. So a fire-weak mob gets 150% of fire damage, plus 100% of lightning damage.
