No one person is able to kill pain unless they were able to take out more than one at the same time. Hanzo lost to pain too, and pretty easily seeing as the flashback showed pain wasn't even scratched.
As for Tsunade healing up the entirety of the village, you have to remember that Kanoha's the largest village out of all of them. And she's not just healing up ninjas, but the citizens too. Tsunade has a lot of chakra + her reserve chakra crystal. If she entered comatose, it means that she nearly depleted all her chakra. If Tsunade were to use her chakra crystal on herself she'd be able to kill, most probably, at least two Pains considering that she's technically immortal in that state. Instead she did what Gaara did and sacrificed herself for her people.
Pain's the strongest member of Akatsuki barring Madara so you can't say that anyone who died against Pain is weak or not deserving of the title of Kage. Pain killed Jiraya, who was also a possible Kage, and Hanzo, who was kage level as the leader of Ame.
