100 def vs 200 def is like 20~30 damage.
The only classes that really see it are warriors and mages. Warriors gears usually add a lot of def, but it's not even that much. Mages see it the most as each int = 1 mdef so they can literally take 100s less of magic damage compared to non-mages
Just go look at the defs -.- It doesn't depend on the character at all really. If they were all equipless, all their defs would read 0 [except mages] and they'd all take the same damage.
