For your first question, I think that giving the letter to the respective NPC will allow you to fight a weaker version of Frankenroid, not entirely sure though.
By the way, I think snipers are best at protecting romeo/juliet at the boss stage because they have pkb, puppet, and rely more on equipment than I/Ls, meaning that they can strip off equips and use the level 12 crossbow so that they would barely deal any damage, preventing new neohuroids from spawning and keeping the ones that are already there immobile. This might also apply to rangers with arrow bomb. I/Ls rely more on base int for their damage, meaning that neohuriods might get killed, allowing them to spawn back, before the rest of the party finishes off frankenroid.
