Note: This has only been verified in the Japanese version of the game. Overseas versions are expected to follow it though. There are some rumours and those are noted through the notes.

This leaves two characters, Wario and R.O.B.. Those are just 30 and 70 VS matches respectively. Should be easy to do by just playing the game.
R.O.B. has a rumour of being unlocked by playing with 6 different unlocked characters in Classic Mode. Wario has no such thing, its alternative lock method is through 100 Man Brawl.
My Confirmations
Dr.Mario does not require any intensity. I was doing all my Classic Mode runs at 4.5 for a long time, and the only three characters I did not unlock without increasing it are Ganondorf, Bowser Jr., and ROB.
Ganondorf and Bowser Jr. require 6.0 or higher intensity. It's the intensity that allows Master Core to appear if you choose to fight both hands at once instead of just Master Hand, however both hands are not required to unlock any character.
I'm not so sure about either of those Jigglypuff requirements. I don't remember what exactly unlocked her for me, but I never used Charizard in Classic Mode. In fact, the only Pokemon I did use was Greninja and I unlocked her after. It may just be a usage requirement, and not Classic Mode. I honestly don't remember.
ROB requires Game and Watch on 6.0 or higher.
